Title: "The role of intermittent and chronic hypoxia in the neuronal control of breathing and swallowing in health and disease"
Our studies focus on understanding the neuronal basis of a variety of brain functions to find novel ways to treat and cure neurological disorders in children, including epilepsy, Rett syndrome, brain tumors, and sudden infant death syndrome (SIDS). Our current work focuses on the analysis of the in vitro respiratory network in mice. For this purpose we isolate acutely the respiratory network in a transverse plane of the mouse medulla. This brainstem slice preparation contains the essential medullary structures involved in cardio-respiratory control and even after the in vitro isolation generates rhythmic activity in rats and mice of all developmental stages (up to an age of 25 postnatal days). This approach permits a direct comparison of the neuronal mechanisms of rhythmogenesis in newborn and more mature mammals. Our experiments indicate e.g. that the hypoxic response, fast chloride mediated inhibitory synaptic transmission, calcium channels and modulatory processes change postnatally within the respiratory network. He is recipient of the International award for Science, Honor and Truth,

Title: "Factors affecting the exercise performance of highland natives in the Andes and Himalayas"
Human biology, high-altitude adaptation, physical activity, growth and development.Professor Brutsaert has broad interests in how gene and environment interact to produce variation in human athletic ability and health and disease. He conducts field research on high altitude natives in the Andes, with some focus on gas exchange and the control of breathing. He and his collaborators have been using genome-wide approaches to elucidate the genetic basis of variation in specific altitude adaptive traits in several Andean populations, including the Quechua, in Peru, and the Aymara, in Bolivia. Brutsaert also has a laboratory-based program that focuses on how early life (intrauterine) developmental effects influence later-life adult exercise capacity, physical activity, body composition, the response to training, and the future risk for chronic disease. Several projects in this area have been conducted with US college-aged students and with rural children in Costa Rica. For the latter, the interest is in whether early life conditions program physical activity patterns and muscle function during childhood. In addition to his research interests, Brutsaert helps to coordinate the Human Performance Laboratory (HPL) at Syracuse University, and is the director of the Altitude Simulation Laboratory (ASL). The ASL houses a human-scale hypoxia chamber that is large enough to accommodate several test subjects and investigators during metabolic and/or exercise testing under simulated altitude conditions up to 20,000 feet. Brutsaert holds a courtesy appointment in the Department of Anthropology and collaborates with faculty at the SUNY Upstate Medical Institute of Human Performance (IHP) which houses clinical facilities and advanced imaging and muscle function technologies.

We use a comparative evolutionary approach in mammals, birds, and fish to understand the mechanistic basis for variation within and between vertebrate groups. Integrative approaches explore the causal links between levels of organization – from gene/genome to physiological system to organism. Some of our ongoing projects are discussed below. Title: "HIF-2 and the carotid body"
My research interests are in understanding the role of hypoxia in cancer. Specifically, we are interested in defining the circumstances and molecular pathways by which hypoxia and hypoxia signalling directly promote cell proliferation. Genetic mutations of hypoxia/HIF signalling pathway components are commonly associated with certain types of cancer e.g. carotid body paragangliomas, adrenal medullary pheochromocytomas, kidney clear cell carcinomas and understanding the mechanisms involved may help with treatment of these cancers. We have shown that Type I cells in the carotid body proliferate in response to hypoxia and that this (like VHL-associated kidney cancer) is dependent on a specific isoform of HIF, HIF-2alpha (Fielding et al., 2018; Hodson et al., 2016). Further, we have shown that unrestrained activation of HIF-2alpha in the mouse carotid body leads to a paraganglioma-like growth (Fielding et al., 2018), mimicking the human genetic condition and providing the first such mouse model to explore the mechanisms and potential therapeutic treatments of this and other HIF-2 dependent cancers.
